Mission

Die Steigerung der Nährstoffnutzungseffizienz ist von entscheidender Bedeutung für eine nachhaltige Pflanzenproduktion und für die Aufrechterhaltung hoher Erträge auch unter ungünstigen Bedingungen, beispielsweise aufgrund des Klimawandels. Um dieser Herausforderung zu entgegnen, konzentriert sich unsere Forschung auf die Aufnahme und den Transport von Mineralstoffen und deren Auswirkungen auf die Pflanzenphysiologie und -entwicklung, mit besonderem Schwerpunkt auf Stickstoff (N), Phosphor (P), Eisen (Fe), Zink (Zn) und Kupfer (Cu) sowie dem nützlichen Element Silizium (Si).

Wir wollen herausfinden, wie von den Nährstoffen abgeleitete Signale die Phytohormon-Signalwege und die nachfolgenden Entwicklungsprozesse modulieren, um die Architektur und Funktion des Wurzelsystems für eine effiziente Nährstoffaufnahme anzupassen. Wir untersuchen auch, wie die von den Wurzeln abgegebenen Metaboliten den Pflanzen helfen, Nährstoffe aus schwer zugänglichen Quellen zu mobilisieren und wie die Nährstoffe effizient durch den Pflanzenkörper und in die Körner transportiert werden. Das Verständnis dieser Prozesse wird dazu beitragen, Nutzpflanzen mit höherem Nährstoffgehalt zu erzeugen, die weniger auf den hohen Einsatz chemischer Düngemittel angewiesen sind. 

Neue kritische Gene und Mechanismen werden durch die Nutzung der genetischen Variation bei Modellpflanzen und in den genetischen Ressourcen der Bundeszentrale Ex-situ-Genbank am IPK entdeckt und erforscht. Die Funktion identifizierter Schlüsselgene wird durch physiologische und molekulare Analysen genom-editierter und transgener Pflanzen in Experimenten charakterisiert, die vom Labor bis zu feldrelevanten Bedingungen reichen. Diese Analysen werden durch unsere analytischen Plattformen unterstützt, die die Messung von Mineralelementen und stabilen Isotopen (ICP-OES, ICP-MS und IR-MS), Phytohormonen (UPLC-MS/MS) sowie Primärmetaboliten und spezifischen Metabolitgruppen (IC-MS und UPLC-MS/MS) umfassen.

Nutrient-dependent regulation of root system architecture

As sessile organisms, plants need to modulate root system architecture to temporal and spatial changes of nutrient availabilities in soils to sustain optimal growth and development. Studies from our research group have identified molecular mechanisms that integrate nutrient cues with root developmental processes to trigger nutrient-specific root architectural changes (e.g., Lima et al., 2010; Giehl et al., 2012; Gruber et al., 2013; Jia et al., 2019; Meier et al., 2020; Jia et al., 2021; Liu et al., 2022; Jia et al., 2023; Maniero et al., 2024).

Our current research involves the identification and characterization of novel molecular players that contribute to nutrient-induced root plasticity. We are also exploring the impact of hormone-related protein variants (proteoforms) in enhancing root responses to limiting nitrogen.  Furthermore, we are using large-scale phenotyping approaches, including the rhizotron system of the IPK PhenoSphere, to investigate the reaction of barley and wheat roots to different nitrogen availabilities. Genome-wide association studies (GWAS), transcriptomics, ionomics, hormonal profiling and genome-editing are used to identify and characterize the underlying genes and to establish which root architectural traits are best associated with nitrogen uptake.

The genes identified in our studies can be exploited in breeding programs to develop crops with root system architecture optimized for more efficient nutrient capture.

Ionomics and nutrient transport mechanisms in plants

Ion transporters located at the plasma-membrane and tonoplast of plant cells mediate the uptake, storage, and translocation of essential and non-essential mineral elements. Consequently, the activity of these transporters and their regulation determine mineral element composition (ionome) of plant tissues. Previous studies of our group have characterized transporters and regulatory mechanisms underlying the uptake and distribution of nitrogen, phosphorus, iron and manganese in plants (e.g., Yuan et al., 2013; Eroglu et al., 2016; Eroglu et al., 2017; Giehl et al., 2017; Duan et al., 2018; Riemer et al., 2021).

Among our ongoing projects, we are investigating the interplay between ammonium and nitrate uptake transporters in A. thaliana. In barley, we are characterizing a novel zinc transporter determining the distribution of this micronutrient in multiple tissues, and screening the nitrogen uptake capacity of genetically diverse genotypes. Furthermore, building on a developed method for cell type-specific ionomics (Giehl et al., 2023), we are investigating nutrient transport pathways at high spatial resolution. All these projects are strongly supported by our element analysis platforms.

The findings of these projects can instruct the development of breeding strategies to enhance nutrient uptake, promote the efficient delivery of nutrients to leaves and grains, and mitigate the accumulation of harmful elements in plant tissues.

Chemical composition and function of root exudates in nutrient acquisition

Root exudates are a complex mix of various metabolites, including amino acids, organic acids, sugars, phenolics and secondary metabolites. They help plants to acquire nutrients by shaping the root-associated microbiota or by directly mobilizing nutrients from sparingly available pools in the soil. Our group has identified key steps in the biosynthesis of the major coumarins secreted by Fe-deficient A. thaliana plants and determined the physiological functions of two coumarins with iron-mobilizing activities (Schmid et al., 2014; Rajniak et al., 2018; Paffrath et al., 2024). We are further exploring the pH-dependent regulation of coumarin biosynthesis in a diverse set of natural accessions.

We are also characterizing the chemical diversity of root exudates in non-model species, such as barley and catch crops (Heuermann et al., 2023). In barley, we are setting special focus on phosphorus deficiency, which is known to induce the synthesis and secretion of various metabolites able to solubilize phosphate from precipitated complexes or to attract symbiotic or beneficial microbial communities. By analyzing genetic resources and genome-edited plants, we aim to identify genes and allelic variants determining the biosynthesis and secretion of metabolites that can improved phosphorus uptake efficiency in crops.    

Approaches to improve abiotic stress tolerance in crops

Environmental stresses impose an increasing challenge for the stability of crop yields in modern agriculture. In the past, we characterized the response of plants to various abiotic stresses, including heavy metal excess and flooding (Lešková et al., 2017; Lešková et al., 2019; Cid et al., 2024). Growth and stress tolerance could be improved with biotechnological approaches, such as the introduction of flavodiirons proteins from cyanobacteria into A. thaliana and barley (Tula et al., 2020; Shahinnia et al., 2021), or by supplying plants with the beneficial element silicon (Beier et al., 2022).

Currently, we are combining physiological and molecular approaches to decipher how silicon improves the tolerance of barley plants to drought. We also investigate the effect of biostimulants in ameliorating the tolerance of tomato and quinoa to salinity, and characterize genotype-specific metabolic adaptations of wheat plants in response to drought. Furthermore, we explore the physiological and molecular mechanisms underlying the plant growth promotion mediated by plasma-activated water.

Bioinformatics @ the Department of Physiology and Cell Biology

Anja Hartmann

In tight collaboration with scientists within and beyond the department, the bioinformatics research activities are mainly focused on the computational analysis of RNA sequencing (RNA-seq) data. The main research objectives are to identify nutrient-responsive gene regulatory networks and infer the functional relationship of genes with changes in physiology, biochemistry and development associated with plant growth, yield formation, and tolerance to biotic and abiotic stresses. To derive these biological insights from RNA-seq data of crops, such as barley and wheat, workflows ranging from raw data processing to intuitive data exploration and interpretation are developed in R utilizing various Bioconductor packages.
