Experimental Taxonomy
Biological taxonomy consists of the sub-areas of nomenclature (use of correct names for organisms), phylogenetics (determination of evolutionary relationships) and systematics (grouping of organisms into meaningful units). The working group deals with questions relating to the reconstruction of relationships, the evolution of characteristic traits and the influence of geological and ecological factors on species stability and speciation. The aim of the research is to understand the mechanisms and processes of speciation (through splitting, hybridisation or chromosome changes), the cohesion of species (under what conditions does the formation of new species not occur?) and the principles of ecological adaptation. Since molecular characteristics play a major role in plant systematics and genetic processes play are important for speciation, there are many cross-connections with genome research.
We employ a wide range of molecular, microscopic and morphological methods. To analyse speciation processes, we use population genetic and phylogeographic approaches together with phylogenetic analyses, chromosome painting, greenhouse experiments, comparative cultivation of species, and field studies in the natural distribution areas of our study objects (currently with a focus on the Mediterranean, the Balkans, and the steppe regions of the northern and southern hemisphere). These are complemented by modelling of the current potential climatic niches of species and their distribution areas during the last ice age. Together with molecular population analyses, these methods allow statements to be made on the age of species and species groups, on possible refugial areas and dispersal routes and on the evolution of morphological and physiological traits through time. The group also curates IPK's herbarium (GAT), a collection of some 500,000 specimens.

Projects
1) Research into the relationships of species and genera within the grass tribe Triticeae
This tribe of about 300 species includes important cereals (barley, rye, wheat). The taxa are mostly distributed in open habitats in temperate regions of the world. Hybrid species and allopolyploids have arisen through repeated crosses between species (also from different genera), making the reconstruction of relationships extremely complicated. By analysing the sequences of approx. 400 genes, which are evenly distributed across the genome, as well as entire chloroplast genomes, it is possible to reconstruct relationships within the Triticeae. Genome-wide SNP data (single mutations) also allow gene flow between closely related species to be recorded. For the closest wild relatives of wheat (Triticum), we were thus able to show that hybridisation not only played a decisive role in the evolution of wheats but also in the origin of the entire genus Aegilops.
2) Detailed analysis of the genus Hordeum (barley)
For the barley genus Hordeum, we are developing a very detailed evolutionary history, ranging from relationships to dispersal events, ecological adaptations, hybridisations and changes in the ploidy levels of the species. By sequencing high quality genomes for all diploid and some polyploid species and analysing them, we can understand the role of genomic changes on speciation, phenotype and ecology.
3) Revision of the genus Crocus
Detailed morphological and molecular analyses of crocuses have increased the number of known species from around 80 to over 200 in the last two decades. We are currently working on creating a new framework for the genus that classifies all species in a system that reflects their evolutionary history and relationships. To elucidate this, we are using sequence information of entire chloroplast genomes, genes of the nuclear genome and genome-wide single mutations (SNPs). In addition to the relationship of the species, we can also estimate their ages from these data and, thus, understand speciation in the context of geological changes.
4) Crocuses have extremely unstable karyotypes
An unusual characteristic of crocuses is the very rapid change in chromosome numbers, sometimes within and often between closely related species. Breakage and/or fusion of (parts of) chromosomes also changes the DNA arrangement within individual chromosomes. We combine cytogenetic methods, in which individual chromosomes are specifically stained, with genome sequencing in an attempt to understand these changes and the underlying mechanisms.
5) The evolution of saffron (Crocus sativus)
The origin of saffron and the saffron crocus has been the subject of speculation for over a hundred years. By analysing the relationship of various crocus species and detailed studies of populations of Crocus cartwrightianus, we were able to clarify the origin of saffron: The spice plant originated in the Attica region of Greece from wild C. cartwrightianus. The coveted triploid, sterile plant then spread through trade and is now cultivated as a single clone in different regions of the world. We are currently working to better understand the differences between the diploid parent and the triploid cultivated form, particularly with regard to the metabolic pathways involved in the production of saffron's constituents. Among other things, we are creating reference genomes for saffron and its wild ancestors and analyse the influence of epigenetic genome modifications on ecological adaptations and important ingredients.
Other projects include the genotyping of the garlic accessions (Allium sativum) of European genebanks and the common onions (Allium cepa) of the IPK genebank, the elucidation of the domestication of coriander (Coriandrum sativum), and taxonomic work in mints (Mentha). Furthermore, we are involved in a DFG priority programme for research into evolution at the dry limit, in which plant environmental adaptations in the Atacama and Namib are analysed. All these projects are part of national or international co-operations.
