Mission

Biologische Taxonomie besteht aus den Teilgebieten Nomenklatur (Verwendung korrekter Namen für Organismen), Phylogenetik (Bestimmung evolutionärer Verwandtschaftsbeziehungen) und Systematik (Gruppierung von Organismen in sinnvolle Einheiten). Die Arbeitsgruppe bearbeitet Fragen zur Rekonstruktion von Verwandtschaftsbeziehungen, Evolution von Merkmalen und zum Einfluss geologischer und ökologischer Faktoren auf Artstabilität und Artbildung. Ziel der Forschung ist es, Mechanismen und Prozesse der Artbildung (durch Aufspaltung, Hybridisierung oder Chromosomenveränderungen), den Zusammenhalt von Arten (Unter welchen Bedingungen kommt es nicht zur Ausbildung neuer Arten?), sowie Grundlagen ökologischer Adaption zu verstehen. Da molekulare Merkmale eine Hauptrolle in der Pflanzensystematik einnehmen und genetische Prozesse bei der Artbildung eine wichtige Rolle spielen, ergeben sich viele Querverbindungen zur Genomforschung.

Wir nutzen eine weite Palette molekularer, mikroskopischer und morphologischer Methoden. Zur Analyse von Artbildungsprozessen verwenden wir populationsgenetische und phylogeographische Ansätze zusammen mit phylogenetischen Analysen, Chromosomenfärbungen, Gewächshausexperimenten, vergleichendem Anbau der Arten, sowie Feldstudien durch Expeditionen in die natürlichen Verbreitungsgebiete unserer Studienobjekte (derzeit mit Schwerpunkten im Mediterraneum, dem Balkan und den Steppengebieten der Nord- und Südhemisphäre). Diese werden ergänzt durch Modellierungen der heutigen potentiellen klimatischen Nischen von Arten sowie deren Verbreitungsgebieten während der letzten Eiszeit. Zusammen mit molekularen Populationsanalysen erlauben diese Methoden Aussagen zum Alter von Arten und Artengruppen, zu möglichen Refugialräumen und Ausbreitungswegen sowie zur Evolution von Merkmalen durch die Zeit. Zur Arbeitsgruppe gehört auch das Herbarium des IPK (GAT) mit seinen ca. 500.000 Belegen.

Projekte

1) Erforschung der Verwandtschaft von Arten und Gattungen innerhalb der Grastribus Triticeae.

Zu dieser Tribus mit ca. 300 Arten gehören wichtige Getreide (Gerste, Roggen, Weizen). Die Taxa sind zumeist in offenen Habitaten der temperaten Regionen der Erde verbreitet. Durch immer wieder vorkommende Kreuzungen zwischen Arten (auch aus verschiedenen Gattungen) entstanden Hybridarten und Allopolyploide, die die Rekonstruktion von Verwandtschaftsbeziehungen extrem kompliziert machen. Durch Analyse der Sequenzen von ca. 400 Genen, die gleichmäßig über das Genom verteilt angeordnet sind, sowie ganzer Chloroplastengenome ist es möglich, die Verwandtschaft innerhalb der Triticeae zu rekonstruieren. Genomweite SNP-Daten (Einzelmutationen) erlauben zusätzlich Genfluss zwischen nah verwandten Arten zu erfassen. Für die nächsten wilden Verwandten des Weizens (Triticum) konnten wir so zeigen, dass Hybridisierung eine entscheidende Rolle in der Entstehung der Gattung Aegilops gespielt hat.

 

2) Detaillierte Analyse der Gattung Hordeum (Gerste)

Für die Gerstengattung Hordeum erarbeiten wir eine sehr detaillierte Evolutionsgeschichte, die von Verwandtschaftsverhältnissen über Ausbreitungsereignisse, ökologischer Anpassungen, Hybridisierungen bis zu Änderungen der Ploidiestufen der Arten reicht. Durch das Sequenzieren von Genomen in hoher Qualität für alle diploiden und einige polyploide Arten und deren Analyse können wir die Rolle genomischer Veränderungen auf Artbildung, Phänotyp und Ökologie erfassen.

 

3) Revision der Gattung Crocus

Durch detaillierte morphologische und molekulare Analysen von Krokussen erhöhte sich die Zahl bekannter Arten in den letzten zwei Jahrzehnten von ungefähr 80 auf nun über 200. Wir arbeiten derzeit daran, für die Gattung eine neue Systematik zu erstellen, die alle Arten in ein System einordnet, das die Entstehungsgeschichte und Verwandtschaftsverhältnisse widerspiegelt. Um diese aufzuklären, benutzen wir Sequenzinformation ganzer Chloroplastengenome, Gene des Kerngenoms sowie genomweiter Einzelmutationen (SNPs). Neben der Verwandtschaft der Arten können wir damit auch deren Alter abschätzen und Artbildung so im Kontext geologischer Veränderungen verstehen.

 

4) Krokusse haben extrem instabile Karyotypen

Eine ungewöhnliche Eigenschaft von Krokussen ist die sehr schnelle Änderung von Chromosomenzahlen, teilweise innerhalb und häufig zwischen nah verwandten Arten. Durch Zerbrechen und/oder Fusion von Chromosomen(teilen) ändert sich ebenfalls die DNA-Anordnung innerhalb einzelner Chromosomen. Wir kombinieren zytogenetische Methoden, bei denen einzelne Chromosomen spezifisch gefärbt werden, mit Genomsequenzierungen und versuchen so diese Änderungen nachzuvollziehen sowie die zugrunde liegenden Mechanismen zu verstehen.

 

5) Die Evolution von Safran (Crocus sativus)

Über die Herkunft von Safran und des Safrankrokus wurde seit über hundert Jahren spekuliert. Durch die Analyse der Verwandtschaft von diversen Krokusarten sowie detaillierte Untersuchungen von Populationen von Crocus cartwrightianus konnten wir den Ursprung von Safran aufklären: Die Gewürzpflanze entstand in der Region Attika in Griechenland aus wildem C. cartwrightianus. Die begehrte triploide, sterile Pflanze verbreitete sich danach durch Handel und wird heute als ein einziger Klon in unterschiedlichen Regionen der Welt angebaut. Derzeit arbeiten wir daran, die Unterschiede zwischen der diploiden Ausgangs- und der triploiden Kulturform besser zu verstehen, besonders im Hinblick auf die Grundlagen der Stoffwechselwege, die in die Entstehung der Inhaltsstoffe von Safran involviert sind. Dazu erstellen wir unter anderem Referenzgenome für Safran sowie dessen wilden Vorfahren und analysieren den Einfluss epigenetischer Genommodifikationen auf ökologische Anpassungen und wichtige Inhaltsstoffe.

 

Weitere Projekte beinhalten die Genotypisierung der Knoblauch Akzessionen (Allium sativum) europäischer Genbanken und der Küchenzwiebeln (Allium cepa) der IPK Genbank, die Aufklärung der Domestikation von Koriander (Coriandrum sativum), und taxonomische Arbeiten in Minzen (Mentha). Weiterhin sind wir an einem DFG-Schwerpunktprogramm zur Erforschung von Evolution am Trockenlimit beteiligt, in dem pflanzliche Umweltanpassungen in der Atacama und Namib analysiert werden. Alle diese Arbeiten sind Teile nationaler oder internationaler Kooperationen.
